cyclin H Antibody

Background

Cyclin H regulates CDK7, the catalytic subunit of the CDK- activating kinase (CAK) enzymatic complex. CAK activates the cyclin-associated kinases CDC2/CDK1, CDK2, CDK4 and CDK6 by threonine phosphorylation. CAK complexed to the core-TFIIH basal transcription factor activates RNA polymerase II by serine phosphorylation of the repetitive carboxyl-terminus domain (CTD) of its large subunit (POLR2A), allowing its escape from the promoter and elongation of the transcripts. It is involved in cell cycle control and in RNA transcription by RNA polymerase II. Its expression and activity are constant throughout the cell cycle.

Purification

Rabbit IgG is purified by Protein A/G affinity chromatography method.

Please use anti-rabbit IgG secondary antibodies.

Immunogen

cyclin H antibody was produced in rabbits immunized with a synthetic peptide corresponding to a region of human CCNH.

Application

cyclin H antibody can be used for Western blot at a suggested dilution of 1 μg/ml in 5% skim milk / PBS buffer, and HRP conjugated anti Rabbit IgG should be diluted in 1: 50,000 - 100,000 as secondary antibody. The protein size is 323 amino acids.

Buffer

Antibody is supplied in PBS buffer with 2% sucrose. Product may ship lyophilized. To reconstitute add 100μl of distilled water making the final concentration 1 mg/ml.

Storage

cyclin H antibody is supplied in PBS buffer containing 0.02% sodium azide. For longer periods of storage, store at -20ºC. Avoid repeat freeze-thaw cycle.
