Need a quick approach to find a good polyclonal antibody, and don’t have recombinant protein available? Producing polyclonal
antibodies using several peptides can be a good solution. However, separate immunization protocols can become very expensive, very
quickly. ProSci offers packages for synthesizing, conjugating, and immunizing multiple peptides into one set of animals. Benefits
of peptide cocktail antibody production include:
- Ability to characterize four or six antibodies to one protein (antibodies against your choice of two or three peptides in two
animals)
- Higher likelihood of finding antibodies that work for multiple applications, particularly epitope-quantity specific applications
such as immunoprecipitation
- Lower cost than separate protocols
Peptide Cocktail Antibody Production Packages
ProSci offers four peptide cocktail antibody production packages, each of which provides a minimum of four antibodies against
your protein of interest.
Package 2PC2 (2 Peptide Package 2):
- Includes separate synthesis, conjugation, immunization and ELISA of two (2) peptides into one set of animals. 5 mg of each
peptide is provided to the researcher, so other assays or purification column construction can be performed in your own lab.
- 4 antibodies can be characterized, two peptide antibodies in each of two rabbits
Package 2PC3 (3 Peptide Package 2):
- Includes separate synthesis, conjugation, immunization and ELISA of three (3) peptides into one set of animals. 5 mg of each
peptide is provided to the researcher, so other assays or purification column construction can be performed in your own lab
- 6 antibodies can be characterized, three peptide antibodies in each of two rabbits
Package 3PC2 (2 Peptide Package 3):
- Includes separate synthesis, conjugation, immunization, ELISA and immuno-affinity purification of two (2) peptides into one set
of animals. 5 mg of each peptide is provided to the researcher, so other assays can be performed in your own lab
- 4 antibodies are purified and can be characterized, two peptide antibodies in each of two rabbits
Package 3PC3 (3 Peptide Package 3):
- Includes separate synthesis, conjugation, immunization, ELISA and immuno-affinity purification of three (3) peptides into one
set of animals. 5 mg of each peptide is provided to the researcher, so other assays can be performed in your own lab
- 6 antibodies are purified and can be characterized, three peptide antibodies in each of two rabbits
Peptide Cocktail Antibody Production Purifications:
Purifying antibodies against specific peptides in a peptide cocktail needs an extra step when compared with traditional peptide
antibody purification. Each animal’s serum contains a pool of antibodies against each peptide sequence; to isolate each
peptide-specific antibody ProSci’s packages that include purification (3PC2 and 3PC3) include the separate purification of
antibodies from each animal, and against each peptide. For example, for package 3PC3 (3 peptide package 3), two animals (animals
1 and 2) would be immunized with peptides A and B. ProSci’s purification procedures yield antibodies for:
- 1A (antibodies from Rabbit 1 against peptide A)
- 2A (antibodies from Rabbit 2 against peptide A)
- 1B (antibodies from Rabbit 1 against peptide B)
- 2B (antibodies from Rabbit 2 against peptide B)
- 1C (antibodies from Rabbit 1 against peptide C)
- 2C (antibodies from Rabbit 2 against peptide C)
Frequently Asked Peptide Cocktail Antibody Production Questions:
How long can the peptides be?
15 residues are included for each separate peptide in the packages, but you can add additional residues for $20 each, up to
35 residues per peptide.
What length of peptides do your recommend for peptide cocktail antigens?
The length for peptides used in this doesn’t need to be as long as for single peptide antigens, but can range from
~12-25 residues.
What purity are the peptides you synthesize?
For peptide cocktail antigens we synthesize for >70% purity, although our average purity is 85%.
Can you use more peptides than three for one project?
At this time we only offer up to three peptides into one set of animals.
Do you conjugate each peptide separately?
Yes, we conjugate each peptide to the carrier protein separately. We find this provides a higher quality antigen than
combining multiple peptides into one conjugation procedure.
Will I receive ELISA data against each peptide?
Yes, each animal’s response to each peptide is measured separately and you will receive data on each of these.
How much peptide will I receive with my bleeds?
You will receive 5 mg of each peptide synthesized with your first shipment of antigen. It will be lyophilized, and not in
solution.
