Custom Polyclonal Antibody Production:
Polyclonal antibodies are one of the most widely used research reagents. They can be used for a range of research applications,
including:
- western blotting
- immunohistochemistry
- immunocytochemistry
- flow cytometry
- immunoprecipitation
- neutralization assays
- ELISAs
We offer antibody production packages that cover all scales of projects, from a single project for a researcher targeting one protein,
to the long-term large scale production needed for diagnostics. Animals are placed on an initial immunization protocol; at the end of
the period you have a three day screening period to evaluate the final scheduled bleeds. Animals can then be terminated at no
additional cost, exsanguinated to harvest additional serum (~60mL per rabbit; $50), or placed on an extension schedule for
longer-term production ($75 per month per animal for rabbit and chicken, $200 per month per animal for goat). Animals can be maintained on extension schedules for
multiple years, allowing you to obtain as much anti-serum as is needed for your research.
Custom Polyclonal Antibody Production Packages:
We offer custom antibody production packages for client-provided antigens, single-peptide projects, peptide cocktail projects,
and phospho-peptide projects.
Custom Antibody Production Package 1 uses a client-provided antigen, and includes an 8-week
immunization protocol in a species of your choice. We can start a project with as little as 0.25mg of your antigen, or perform a
complete immunization protocol with as little as 0.5mg of antigen in two chickens. We can perform additional services (ELISAs,
purifications, western blotting, etc.) for any project to assist you in evaluating the antibodies produced.
Custom Antibody Production Package 2 includes complimentary antigen design, synthesis of a
15-residue peptide at >70% HPLC purity, conjugation of the peptide to a carrier protein, an 8-week immunization protocol in a species
of your choice, and an ELISA to verify titers on the first production bleed; a titer of >1:5,000 is guaranteed if you use a peptide
sequence we approved.
Custom Antibody Production Package 3 includes complimentary antigen design, synthesis of a
15-residue peptide at >70% HPLC purity, conjugation of the peptide to a carrier protein, an 8-week immunization protocol in a species
of your choice, an ELISA to verify titers on the first production bleed, construction of an immuno-affinity column using project
peptide, and purification of 25mL of serum or yolk; a titer of >1:5,000 is guaranteed if you use a peptide sequence we approved.
Custom Antibody Production Package 4 includes complimentary antigen design, synthesis of a
15-residue peptide at >70% HPLC purity, conjugation of the peptide to a carrier protein, an 8-week immunization protocol in a species
of your choice, an ELISA to verify titers on the first production bleed, construction of an immuno-affinity column using project
peptide, and purification of 200mL of serum or yolk; a titer of >1:5,000 is guaranteed if you use a peptide sequence we approved.
Peptide Cocktail Antibody Production Packages include all of the services in Package 2
and Package 3 respectively, but include the synthesis, conjugation, and immunization of your choice of 2 or 3 peptides into one set of
animals. This is an option that increases the likelihood of a peptide-based antibody working for multiple applications, as it doubles
or triples the available number of epitopes for the animals’ immune system to respond against. It also helps to reduce overall cost,
as it minimizes the number of animals needed for a particular project.
Phospho-Specific Antibody Production Packages include all of the services in Package 2,
3, and 4 respectively, but include the synthesis, conjugation, and immunization of phospho peptides into a set of rabbits. Packages
2PN, 3PN, and 4PN also offer the synthesis of the corresponding non-phospho peptide sequence, used for isolating both the phospho
and non-phospho antibodies for your target protein.

Custom Phospho-Specific Antibody Production:
The most common peptide modification used for custom polyclonal antibody production is the phosphorylation of amino acids. This
is accomplished immunizing host animals with a short synthetic peptide containing the phosphoryl (PO3)
group on the residue of interest. The generation of custom phospho-specific antibodies can ensure that the peptide mimics the natural
conformation and activity found in the protein. Phosphorylation often results in functional changes in the activity of a protein,
and having antibodies recognizing the phosphorylated state (as well as the corresponding non-phosphorylated state) can be integral
for studying protein pathways.
There are three residues which can be phosphorylated:
- Serine (S)
- Threonine (T)
- Tyrosine (Y)
The process of producing custom antibodies against a peptides containing a phospho-residue is a bit more complicated than traditional
peptide antibody production. The animals can produce antibodies against additional epitopes in the peptide sequence, as well as
produce antibodies that do not contain the modified phospho-residue. While a pool of phospho and non-phospho antibodies can be
difficult to work with, if purified from the anti-serum the non-phospho antibody can be very helpful for studying protein pathways.

Peptide Cocktail Custom Antibody Production:
Need a quick approach to find a good polyclonal antibody, and don’t have recombinant protein available? Producing custom polyclonal
antibodies using several peptides can be a good solution. However, separate immunization protocols can become very expensive, very
quickly. ProSci offers packages for synthesizing, conjugating, and immunizing multiple peptides into one set of animals. Benefits of
peptide cocktail custom antibody production include:
- Ability to characterize four or six antibodies to one protein (antibodies against your choice of two or three peptides in two
animals)
- Higher likelihood of finding antibodies that work for multiple applications, particularly epitope-quantity specific applications
such as immunoprecipitation
- Lower cost than separate protocols
ProSci offers four peptide cocktail custom antibody production packages, each of which provides a minimum of four antibodies against
your protein of interest.
Custom Monoclonal Antibody Production:
ProSci can help you develop the monoclonal antibodies needed for your research. We understand researchers have varying needs for
levels of involvement, depending on the target. We have developed packages for different levels of involvement in the project,
from researcher screening (you screen positive wells following fusion for clones of interest) to ProSci sending western blot and
immunocytochemistry positive final clones. Additionally, ProSci offers packages for protein and peptide antigens.
ProSci’s fusion success rate for hybridoma production is greater than 95%, using techniques developed in-house. All work
is performed by skilled technicians on premises in our Poway, California laboratories. Monoclonal antibody production packages
can be scaled up to almost any magnitude. We can screen against multiple antigens (additional charges apply) to develop the specific
clone you need. ProSci has repeatedly proven our abilities to produce monoclonal antibodies against difficult targets.
We use 5 Balb/c mice as a production strain for monoclonal antibody development. Balb/c is a very common species used throughout the
industry for monoclonal antibody development due to their large spleen to body size ratio. This yields a higher number of
spleenocytes at fusion.
Each of our monoclonal antibody production packages includes:
- Immunization of antigen into 5 Balb/c mice
- Screening of test bleeds against immunizing antigen to isolate a mouse with high IgG titer and low IgM titer
- Fusion of one mouse’s spleen to the SP 2/0 myeloma cell line
- Screening of positive wells following fusion for IgG specific clones using immunizing antigen
- Cell line stabilization of up to 3 cell lines
- Isotype report on each final clone provided, confirming Ig class
- Cell line banking for 1 year following final clone shipment
Ascites Production:
ProSci can produce antibody from a Balb/c-derived cell line through the production of ascites fluid. This is done by injecting the
cell line into the primed peritoneal cavity of Balb/c mice, and harvesting the fluid. We can do this from a cell line produced through
our hybridoma development programs, or from a cell line you send to us.
Our ascites production includes:
- Mycoplasma screening
- Cell line amplification
- Injection of cells into primed Balb/c mice
- 2 taps to obtain ascites fluid
