Custom Antibody Production Species:



Deciding which type of custom antibody project is right for your research needs is the first step towards initiating a project. There are many factors that should be taken into account, including antibody quantity requirements, desired applications/usages, and cross-reactivity.

There are two main types of projects, monoclonal and polyclonal. Each project type has its own set of advantages and disadvantages. The first factor to determine is which species of host animal should be used for the production of the antibodies. The chart below can be used to highlight the benefits of each type of project,depending on your research needs.

Polyclonal Antibodies

Monoclonal Antibodies

Pros: Cons: Pros: Cons:
  • Quick turnaround
  • Inexpensive
  • One antibody can work for a variety of applications
  • Requires less than 1mg immunogen
  • Multiple host species available, including nonmammalian hosts
  • Quantity available limited by number of bleeds
  • Lot-to-lot variability in characterization
  • Difficulty in reproducing a specific antibody in different animals at later dates
  • Unlimited quantity and availability of antibody
  • Lot-to-lot consistency
  • High specificity of antibodies
  • Quick reproduction of antibody through ascites production
  • Longer production time
  • Requires a minimum of 4 mgs immunogen
  • High cost of development
  • Antibodies less likely to work for multiple applications
  • More difficult if using peptide immunogens
Frequent Uses: Frequent Uses:
  • Smaller scale research (needing < 50mgs antibody)
  • Limited protein immunogen availability
  • Rush projects (antibody available in as little as 35 days)
  • Diagnostics
  • Therapeutics
  • Larger scale or long-term research (needing 100+ mgs antibody)


Polyclonal Antibodies:

Species Availability

Chicken

Pros:

  • Non-mammalian host species
  • Good alternative for highly conserved mammalian proteins
  • Non-invasive (antibodies are purified from egg yolks)

Cons:

  • Not ideal for peptide antigens

Ideal Package: 1
(using a highly conserved mammalian protein provided by researcher)

Rabbit

Pros:

  • Most common host species
  • Great host for peptide and most protein antigens
  • Serum can be used directly for initial assays
  • Purifications are rapid and straight forward
  • Antibodies can be delivered in 35 days, using accelerated immunization protocol

Cons:

  • Not ideal for highly conserved mammalian recombinant proteins

Ideal Package: 1, 2, 3, or 4
(using a peptide or a protein for large scale research)

Goat

Pros:

  • Large mammalian host species
  • High quantity (250mL) bleeds for large lots

Cons:

  • Not ideal for modified peptide antigens

Ideal Package: 1, 2, 3, or 4
(using a peptide or a protein for large scale research)

 

Monoclonal Antibodies:

Species Availability

Mouse

  • Most common host species (Balb/c mouse)
  • 5 mice per project gives pool of animals to choose between
  • Easy to produce antibody through either cell culture or ascites production
